Neisseria gonorrhoeae is a gram-negative diplococcus that causes gonorrhea, a sexually transmitted disease (STD) that can cause ectopic pregnancy and infertility, and can increase the risk of contracting and transmitting HIV.
WHO has estimated that 82.4 million new cases were reported in 2020. An estimated 550,000 drug-resistant Neisseria gonorrhoeae infections are detected each year in the United States.
In particular, concerns have been raised about the emergence of Neisseria gonorrhoeae strains resistant to extended-spectrum cephalosporins (ESAs), such as ceftriaxone, which is one of the most effective antibiotics for treating gonorrhoea. Resistance to other antibiotics used in the treatment of gonorrhoea, such as fluoroquinolones and macrolides, has also been reported. Multiple resistance to different classes of antibiotics makes effective treatment of the infection even more difficult.
